Lab Study Explores How Omega-3 and Omega-6 Fats Affect Liver Cell Fat Buildup and Inflammation

This laboratory study used human liver cells (HepG2) to explore how two types of polyunsaturated fats—omega-6 linoleic acid and omega-3 alpha-linolenic acid—interact with a liver signaling system involving an enzyme called butyrylcholinesterase (BChE). The researchers were interested in understanding cellular processes relevant to metabolic dysfunction-associated steatotic liver disease (MASLD), a condition involving fat buildup and inflammation in the liver.

Cells exposed to alpha-linolenic acid (an omega-3 fat) showed early changes suggesting reduced fat production and increased fat-burning activity within the first 24 hours, though these effects lessened by 48 hours. In contrast, linoleic acid (an omega-6 fat) showed a more complex pattern—mild early effects followed by a marked increase in inflammatory markers at 48 hours.

The study also found that overexpressing the BChE enzyme increased genes tied to fat and triglyceride production, and that combining PUFAs with BChE overexpression produced varying effects depending on fat type and exposure time. Additional computer modeling suggested that alpha-linolenic acid could physically interact with certain nerve-signaling receptors in liver cells, hinting at a possible link between cholinergic (nerve-related) signaling and liver fat metabolism.
