Omega-3 Phospholipids May Support Gut Barrier Function During Inflammatory Stress in Mice
This study looked at how DHA-enriched and EPA-enriched phospholipids affected the intestinal lining in mice exposed to a bacterial toxin (lipopolysaccharide, or LPS) that is commonly used to trigger inflammation and gut barrier damage in research settings.
Mice were fed a diet containing 1% DHA-phospholipids or EPA-phospholipids for four weeks before being given an LPS injection. Researchers then examined the intestinal tissue for signs of damage, including cell death (apoptosis), a cellular cleanup process called autophagy, and the integrity of tight junctions that hold intestinal cells together.
Mice pretreated with either DHA-PL or EPA-PL showed better balance between cell death and autophagy, along with more intact tight junction structures compared to untreated mice. When researchers blocked a protein called sirtuin 1 (SIRT1) using a specific inhibitor, the protective effects of both phospholipid types disappeared. This suggests the benefits were tied to SIRT1 activity, which in turn appeared to dampen two inflammatory signaling pathways (NF-κB and MAPKs) while boosting an antioxidant pathway (Nrf2).
What this means for you
This is a mouse study, not a human trial, so it cannot tell us how DHA or EPA phospholipids affect the human gut lining. It does offer early mechanistic clues about how these omega-3 phospholipid forms might interact with cellular stress-response pathways involved in gut barrier integrity, which could inform future human research.
