Seminal Fluid Chemistry May Reveal Signs of Testicular Cancer

Researchers analyzed seminal plasma samples from 37 men with testicular germ cell tumors (TGCTs) and 11 men without this cancer, looking at two groups of molecules: small polar metabolites (using capillary electrophoresis-mass spectrometry) and oxylipins, which are lipid-derived signaling molecules (using liquid chromatography-tandem mass spectrometry).

The metabolite analysis showed disrupted energy metabolism patterns in men with TGCT, including lower levels of certain energy-related amino acids, suggesting the tumor cells had higher metabolic demands. Changes in sugar-related metabolic pathways also hinted at altered protein processing and secretory activity in the reproductive tract.

On the oxylipin side, four specific lipid mediators (9,10-DiHOME, 6-trans-LTB4, TXB2, and 14,15-DiHETrE) were found at higher levels in the TGCT group and, when combined into a ratio, showed strong ability to distinguish cancer samples from controls in this small study. Tissue staining confirmed that an enzyme called ALOX12, involved in producing these lipid mediators, was expressed in the tumor tissue itself, supporting a biological link between the tumor and the oxylipin changes seen in seminal fluid.
