Comparing Omega-3 + Inositol vs. N-Acetylcysteine for Mood Swings and Depression in Kids
This 6-week study looked at children and teens (ages 5-17) who struggled with emotional dysregulation, including some with traits of autism. Researchers compared two nutraceutical approaches: a combination of omega-3 fatty acids (fish oil, providing EPA) plus inositol, versus N-acetylcysteine (NAC), an amino acid derivative.
Both treatments led to modest improvements in symptoms of depression and mood swings (mania), based on ratings from clinicians and parents. Overall, the two treatments did not differ significantly from each other. However, when researchers looked within each group separately, the omega-3 plus inositol group showed a statistically significant improvement in depression scores, while NAC did not reach that same significance for depression.
For mood swings, NAC showed a larger (though not statistically significant) improvement compared to omega-3 plus inositol. Interestingly, in children who also had autism traits, NAC appeared more effective specifically for mood swings, and also showed improvement in measures of social responsiveness and behavioral flexibility that omega-3 plus inositol did not.

This small trial suggests that omega-3 fatty acids combined with inositol may offer some support for mood-related symptoms in children with emotional dysregulation, with a possible edge for depressive symptoms specifically, while NAC showed some advantages for mood swings, especially in children with autism traits.
