Scientists Engineer Gut Bacteria to Produce an Anti-Inflammatory Omega-3 Derivative
Resolvin E1 (RvE1) is a molecule the body naturally makes from omega-3 fatty acids like EPA, and it plays a role in helping inflammation settle down. However, RvE1 breaks down quickly and is expensive to manufacture, which has limited its use in research and clinical settings.
To get around these obstacles, researchers modified a well-studied probiotic strain of E. coli (Nissle 1917) to carry two enzymes, COX2 and 5-LOX, that convert EPA into RvE1 inside the gut. In lab tests, this engineered probiotic successfully converted EPA to RvE1 and was able to persist in the intestines of mice for an extended period.
When tested in mouse models of acute inflammation and colitis (an inflammatory bowel condition), the engineered bacteria were associated with reduced inflammatory markers, protection of gut tissue, and improvements in the balance of gut bacteria. The researchers also showed that a photosynthetic organism called Euglena gracilis could serve as an alternative, plant-based source of the omega-3 fatty acids needed to feed this process.
